I don’t know Manga studio specifically, but in my experience with vector draw programs you can combine vectors into an object and then give that object an interior color.
I would have thought so, too, but MS wouldn’t allow it (could’ve been doing something else wrong, I guess). I’ll have to try it in Inkscape.
